Advantages of small freeze dryer method over traditional drying

Methods for drying are various, sun-drying, cooking, spray-drying, vacuum drying, etc. However, ordinary drying methods are usually performed at a temperature of 0°C or higher. Generally, the products obtained by drying have the problems of shrinking in size and hardening the texture. Most of the volatile components will be lost, some heat-sensitive substances will be denatured and deactivated, and some substances will even be oxidized. Therefore, compared with the product before drying, the properties of the product after drying are very different.

The lyophilization rule is basically carried out below 0°C, that is, in a frozen state of the product, and generally does not exceed 60°C when analyzing and drying. Under vacuum conditions, when water vapor directly sublimates, the drug remains in the ice shelf when it freezes, forming a sponge-like loose porous structure, so its volume will not change after drying. Development of Flash Column Method

 The flash column method is an alternative to long column chromatography developed in 1978.

The method aims to separate the components from the mixture, thereby purifying it. It is established from the existing long-column chromatography technology, which is time-consuming and often unsatisfactory. In short, the flash column method allows the sample to pass through a column filled with gel, and the gel separates the sample.

Still, the founder of the flash column and his colleagues have been using medium pressure chromatography and short column chromatography to replace long column chromatography. They decided to combine the two to overcome the shortcomings of long column chromatography, which is time-consuming and low recovery.

The gel originally used to pack the column was silica gel, which is still widely used. They used air pressure to push the solvent through the silica column, compressing the column. The sample is then applied and the same (can be different, but usually the same) solvent is used to pass the sample through the column. Then collect the purified components or fractions, the whole process takes about 5-10 minutes. Usually, a small part appears first and most eluted last. The collected fractions were subjected to original analysis by thin layer chromatography (TLC) plates.

Since its introduction, the silica gel flash column has been widely used in organic chemistry. However, guidelines are often loose, driven by personal experience, or not translated well when settings are changed. However, some things are still true. Increasing the sample size will cause the resolution to decrease.

Compared with the HPLC column, the resolution of the flash column is already at an intermediate level, but it is sufficient for sufficient separation, so the increase in the number will only worsen the situation. Second, the optimal flow rate depends on the length and width of the column and the nature of the gel.

This is due to the number of plates available, for example, longer and narrower columns will provide more theoretical plates, thereby affecting the flow rate. Finally, the resolution is affected by the stationary phase. If the stationary phase or gel arranged on the column is more uniform and has a smaller particle size, it provides better resolution. The smaller the particle size, the larger the surface area, and the higher the resolution.

Manipulating all these factors to optimize the purity or recovery of the components can be quite complicated because they interact, but they have different effects when tested independently. For example, the mobile phase selectivity has the greatest impact on the resolution, but it depends on the column capacity. Then, this will be affected by the chosen solvent.

Therefore, before starting any real experiment, you need to test or calculate the settings used in the flash column. If you have data from TLC, you can calculate the best settings without testing any flash column settings. Low TLC delay factor (Rf) provides better separation. Using such data, the amount of solvent required can be calculated because they are inversely proportional to each other, and both are related to the column void volume (the solvent in the column before the sample is loaded).

In the original method, it was very important to prevent the column from drying out. However, a more user-friendly alternative has since been developed, called dry column flash chromatography. This method has been adopted by first-time students, while still producing good results comparable to the quality of TLC analysis.

The principle is the same, but the column contains dry silica gel. The powdered xerogel is filled into the column by suction, and finally, a uniform bed of about 1 cm for solvent and sample is obtained. The column was also eluted by suction and dried after each fraction.

Hawach glass solvent filter is designed for solvent particulate removal and aqueous mobile phase filtration. All parts of glass solvent filter contacting with solvents are made of chemically inert glass or PTFE material, which can be used in conjunction with regenerated cellulose (RC) filter membrane, especially suitable for particle removal and degassing filtration of the mobile phase during chemical analysis such as HPLC、GC、AA.

The outlet position is far lower than the vacuum pump interface position, which makes the filtration process safer. A hose made of polypropylene is used to safely connect the vacuum hose.

The function of Vacuum Pump

Hawach vacuum pump is a device that uses mechanical, physical, or chemical methods to obtain a vacuum. Generally speaking, a vacuum pump is a device to improve, produce, and maintain a vacuum in a closed space by various methods.

If the material contains water or organic solvent, add liquid water filter and condenser to play a filtering role. While using a vacuum dryer, we must strengthen the maintenance of the vacuum pump, change the oil to a vacuum pump in time, and prolong its service life.

Structure of Vacuum Filter

The overall structure of the pump is determined by the layout structure of the pump body. The vertical structure of the inlet and outlet horizontal setting, assembly, and connection lines are more convenient.

However, the center of gravity of the pump is high and the stability is poor at high speed, so this type is mostly used for small pumps. The air inlet of the horizontal pump is up and the exhaust port is down. Sometimes for the vacuum system pipe installation and the connection is convenient, the exhaust port can be connected from the horizontal direction, that is, the inlet and exhaust direction is perpendicular to each other.

About The Features And Usages Of Glass Fiber Extraction Thimbles

HAWACH provides customers with two main kinds of extraction thimbles, cellulose, and glass fiber. Today's focus is on the glass fiber ones. The glass fiber extraction thimble belongs to the deep layer filtration, and its main purpose is to be used as the filter layer and directly added to the filter membrane. It is made of 100% glass fiber, with the characteristics of a high flow rate, strong dirt holding capacity, which can filter fine particles. Compared with other fiber materials and synthetic materials, glass fiber extraction thimble has higher filtration efficiency and higher dirt holding capacity.

The HAWACH glass fiber extraction thimbles can be used in Soxhlet extraction: when the temperature is higher than 120℃, strong acid and other cellulose filter cartridges cannot be tried. Soxhlet extractor is used in the determination of crude fat content in commercial grains and oilseeds, whose three major components are heating extraction, solvent recovery, and cooling. The heating temperature of the Soxhlet extractor can be flexibly adjusted according to the ambient temperature during operation and the boiling point of the reagent.

In order to achieve the purpose of rapid extraction, the sample of the fat analyzer is soaked and extracted repeatedly in the extraction process. The solvent can be recovered automatically, which saves a lot of time and greatly facilitates the use of users.  The extraction time is adjustable, and an alarm will be issued when the time comes.

Also, the glass fiber extraction thimbles are to collect particulate matter such as smog in the atmosphere. The effect is better when used with smoke collection equipment. It is suitable for stationary pollution source detection: smoke, and dust, such as cadmium, nickel, tin, fluoride, etc. from fixed atmospheric pollution sources; Fixed pollution source waste gas benzoic ratio, lead, pitch smoke, sulfuric acid fog, etc.; Ambient air and exhaust arsenic, dioxins, etc.
